Colonial Office 43 consists of entry books (copies of out-letters) with four basic internal divisions: volumes 1-15, Quebec, 1763-1801; volumes 16-36, Lower Canada 1791-1842; volumes 37-48, Upper Canada, 1791-1841; volumes 49-158, North America, General, 1810-1873. Colonial Office 43 includes numerous documents dated up to 1782 of the old Board of Trade, particularly in the first four volumes. Included are letters, representations, reports with commissions and instructions sent from the offices of the Board of Trade, 1763-1784, and from the Secretary of State, 1784-1873, usually signed by the Under Secretaries. The class follows the usual system of arrangement for "colony" classes and is available in its entirety on microfilm reels B-831, B-838 to B-868, B-3042 to B-3058. Transcripts are available on reels C-13607 to C-13614.

Notwithstanding the four basic internal divisions mentioned above, volumes 4, 10, and 11 include material relating to both Upper and Lower Canada, despite being in the general "Quebec" category. Within the basic internal divisions, further distinctive units may be identified: volumes 16-21, 38-39 provide précis books for Lower (1791-1814) and Upper (1791-1807) Canada respectively. The précis books, which often digest both in and out-letters as well as action taken, do not readily reveal their purpose, but they are not registers. In many instances, the more complete draft of the communication found in the précis books may be located in other volumes of CO 43.

Volumes 49-141 (1810-1872) include the so-called "Domestic Correspondence", that is, "Offices" and "Individuals" categories, for Nova Scotia, New Brunswick, Newfoundland, Prince Edward Island, British Columbia and Vancouver Island.
